چکیده

Introduction. Human peripheral blood mononuclear cells (PBMCs) are a pool of immune and they also convenient model system for studying pathologies. Aim. Testing bioactivity glycerolipid preparations from fern horsetail species containing long chain polyunsaturated fatty acids (LCPUFAs) towards PBMCs without exogenous stimulation after phorbol-12-myristate-13-acetate (PMA) plus ionomycin stimulation. Materials methods. Glycerolipid preparations were produced by fractionation total lipids, isolated young fronds the Matteuccia struthiopteris shoots Equisetum arvense , on silica. Egg phosphatidylcholine was used comparison. Fatty analyzed gas chromatography. Mononuclear patients with asthma. Parameters cell viability activation estimated flow cytometry. Results. The preparations found to have cytotoxic effect while egg not. most active fraction lipids eluted methanol which reduced 64.6 (51.1-79.0)% in concentration 2 pg/ml caused complete death 20 pg/ml. After PMA/ionomycin, cytotoxic preparation increased although level expressing marker CD69 did not change. other observed higher concentrations (20 and/or 80 pg/ml) it less pronounced: 7.1 (6.7-9.4)% mixture chloroform - water (3:5:2), 39.8 (26.4-41.6)% 12.0 (10.0-15.5)% fractions chloroform-methanol-water mixture, respectively, Conclusion. Comparison acid composition confirm contribution LCPUFAs effects. Identification an component may allow development drug local application hyperimmune response or experiments.

Optimization of human peripheral blood mononuclear cells (PBMCs) cryopreservation

Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...

Toxic Effect of Verapamil on Human Peripheral Blood Mononuclear Cells and BALB/c Peritoneal Macrophages, in vitro

Background & Aims: Verapamil as a calcium channel blocker has been broadly used in the treatment of many cardiovascular diseases such as hypertension and arrhythmia. Furthermore, the anti-tumor/ antiinflammatory effects of verapamil have been shown. In the present study, the cytotoxic effect of verapamil on human peripheral blood mononuclear cells (PBMCs) and BALB/c peritoneal macrophges has be...

Effect of Myeloma-Derived Microparticles on in Vitro Proliferation and Viability of Alloimmune Human Peripheral Blood Mononuclear Cells

Background: Microparticles (MPs) are small vesicles released from the cell membrane. Accordingly, these contain active molecules to mediate biological processes, including cell proliferation and cell cycle progression. The fusion of myeloma cell lines with immunized B lymphocytes is a critical step of hybridoma technology. MPs modulate lymphoproliferation, thereby facilitating B cell expansion ...

Effect of 900 MHz Electromagnetic Radiation on the Induction of ROS in Human Peripheral Blood Mononuclear Cells

Background, Despite numerous studies over a decade, it still remains controversial about the biological effects of RF EMF emitted by mobile phone telephony. Objective, Here we investigated the effect of 900 MHz GSM on the induction of oxidative stress and the level of intracellular reactive oxygen species (ROS) in human mononuclear cells, monocytes and lymphocytes as defence system cells. Metho...

The Influence of Iron Loading and Iron Chelation on the Proliferation and Telomerase Activity of Human Peripheral Blood Mononuclear Cells

Background: Iron is an essential trace element in cell proliferation. Several investigations demonstrate that iron deprivation inhibits cell proliferation. However, the impact of iron on telomerase activity of activated lymphocytes remains unexplained to date. Objective: In this study, the effect of iron on the proliferation and telomerase activity of lymphocytes stimulated by phytohemagglutini...
